Aortic dissection is a highly lethal condition of the major vessels of the heart, with its pathogenesis still not fully understood. This review explores the critical role of transcription factor Runx-related transcription factor-1 (RUNX1) in the pathogenesis of aortic dissection, particularly its involvement in mediating inflammatory responses through various signaling pathways such as nuclear factor kappa-B (NF-κB), interleukin-6 (IL-6), and matrix metalloproteinase-9 (MMP-9). The review covers studies detailing RUNX1's impact on endothelial cell damage, smooth muscle cell phenotypic changes, and extracellular matrix degradation. Potential therapeutic strategies targeting RUNX1 are discussed, highlighting the need for further research to translate these findings into clinical practice.
